Qingdao Institute of Energy, Li Welfare Research Group reveals the fermentation product of lignocellulosic butanol

Using lignocellulose as raw material, the production of liquid biofuels can fundamentally solve greenhouse gas emissions and alleviate the global energy crisis. However, in the bio-fermentation process using corn stalks as a raw material, the pre-treatment of corn stalks causes the lignin to degrade and produce a series of structurally different, soluble compounds called soluble lignin compounds (SLCs). It inhibits the growth of Clostridium butyricum. In this study, in-depth study of the effect of corn straw hydrolysate on Clostridium butyricum was conducted. It was found that corn straw hydrolysate can inhibit the growth and utilization of Clostridium. This inhibition can be removed by calcium hydroxide detoxification.

This study found that SLCs change the acetone/butanol ratio (A/B) during butanol fermentation. The classical A/B fermentation of glucose with a carbon source of Clostridium beijerinckii NCIMB 8052 is about 0.5. In this study, batch fermentation with lignocellulosic hydrolysate as a carbon source was performed. The final A/B was 0.95. Chromatography/mass spectrometry and electrochemical analysis revealed that the soluble lignin in the hydrolyzate can generate oxidative stress. The production of Clostridium butyricum consumes reducing force (NADH) during the removal of oxidative stress. This results in the final fermentation product. Changes in the acetone and butanol ratios resulted in a significant increase in the ratio of acetone in the total solvent. In the process of microbial fermentation, soluble lignin in the hydrolysate can provide electrons and promote the production of hydrogen gas from Clostridium butyricum.

These results show that SLCs not only inhibit the growth of microbial cells, but also adjust the A/B ratio during the fermentation of C. beijerinckii butanol.
